The Petter Brodin’s research group located at Biomedicum, Stockholm is a part of the Unit of Clinical Pediatrics, which is a broad, multi-disciplinary unit at the department of women’s and children’s health, Karolinska Institutet. The lab being a leading labs in systems immunology develops and applies experimental and computational research methods for systems-level analyses of human immune systems.

Description of the subject area

The Brodin lab is a multidisciplinary laboratory investigating human immune system variation and development early in life. The team develops cutting edge technologies for systems-level analyses and apply these to the studies of immune system development in young children and throughout childhood as well as the study of immune dysregulation in the context of disease. The group is half experimental and half computational and our work is cross-disciplinary in nature. The research group coordinates multiple collaborative projects and international collaborations with academic and industry partners.

Your mission

The responsibilities of this candidate in the lab are as follows

• Using cytometry methods for research (conventional flow, spectral flow & mass cytometry) and operation of the instruments with regular maintenance.
• Troubleshooting instruments requiring good understanding of cytometry technologies.
• Antibody panel design and testing
• Custom conjugation of antibodies to metal isotopes that are used as probes in mass cytometry.
• Testing and titrating probes by mass cytometry (and validation by flow cytometry when needed).
• Cell staining and sample multiplexing/barcoding.
• Cytometry data analysis, interpretation, and reports/figures generation.
• Ability to learn newer methodologies/techniques.
• Use automated liquid handling platform.
• Systematic planning of experiments with meticulous approach to recording the observations.
• Setting up immunological assays like – cell stimulation assays, proliferation assays, ELISA, polarization cultures, peptide assays, blood sample prepping etc.

Eligibility requirements

To be eligible for employment as a Research Specialist, in addition to holding a Degree of Doctor, PhD, or having equivalent scientific expertise, the applicant must have demonstrated research expertise as well as have documented research experience after defending their doctoral dissertation. Postdoctoral experience is important.

• Demonstrated experience with a good command and understanding of cytometry (both conventional and spectral flow & mass cytometry) and operation of the instruments with regular maintenance.
• Experience with troubleshooting the machines based on good understanding of the technologies.
• Antibody panel design – knowhow and practical experience of designing panels and the rules to follow based on the cytometry technology in question.
• Conjugation of antibodies to metal isotopes that are used as probes in mass cytometry
• Testing and titrating probes by mass cytometry (and validation by flow cytometry when needed)
• Analytical capabilities of cytometry data using multiple tools/softwares and ability to interpret the data and make reports, contributing to publications etc.
• Ability to learn newer methodologies/techniques.
• Some level of experience in using automated robotic platforms
• Systematic planning of experiments with meticulous approach to recording the observations
• Sound knowledge and experience in setting up immunological assays like - stimulation assays, proliferation assays, ELISA, polarization cultures and other cell cultures, blood sample handling/prepping, peptide assays etc.

The successful candidate must be thriving in interdisciplinary environments and should be dedicated to performing the tasks assigned in the lab error free and with great care. The candidate must be comfortable to work both independently and in collaboration with other team members (when necessary) and contribute to the team’s concerted efforts. The candidate must have good interpersonal skills and communication skills in English as the team is multicultural and interdisciplinary.

• Documented experience of the aforementioned eligibility requirements-
• A research background in immunology.
• Prior lab experience through documented publications and knowledge in biology and cytometry (flow and mass cytometry).
• Motivation letter showing specific interest in the project and job responsibilities.

It is meritorious that the applicant has shown

• Experience of method development with iterative testing of protocols.
• Any computational experience is a benefit.
