Department of Microbiology, Tumor and Cell Biology (MTC) at Karolinska Institutet conducts research and teaching in immunology, infection biology, cell biology and cancer. MTC has about 430 active employees, including 100 PhD students, 50 postdoctoral researchers and 26 professors. Our lab has a long and successful track record of research aimed at discovery of small molecules reactivating p53 and studies of the molecular mechanisms of p53- mediated transcriptional response. We are initiating a new project to dissect the transcriptional regulation of p53 in previously unprecedented detail, using modern methodologies such as CRISPR-Cas9 and high throughput proteomics.

Duties
We are seeking a highly motivated, innovative and skilled Junior Scientist who will perform and supervise the project aimed at elucidation of the mechanisms of p53-mediated gene activation and repression. The project involves the application of high throughput technologies, such as ChIP, transcriptomics and proteomics, as well as CRISPR-deadCas9 applications to identify those genes and p53 cofactors which affect the p53 transcriptional response and which are crucial for p53-dependent biological outcomes.  

Entry requirements
To qualify, the applicant must hold a PhD or has achieved the corresponding scientific competence, and a minimum of 4 years of research experience in relevant fields. The persons appointed should primarily be the ones who have been granted a PhD or have reached the corresponding competence no more than seven years before the end of the application period.

Bases of assessment
A PhD and at least 4 years post-doctoral training in the field of oncogenes and p53 pathways and an extensive experience in protein biochemistry and transcription factors is crucial. Documented experience in relevant research areas as stated below and extensive experience with cell and molecular biology techniques is needed. Experience with project management and supervision of students is also important. Initiative and leadership, strong communication and collaboration skills are also needed.

Extensive experience in the following areas are considered as essential qualifications:

  • Mammalian cell culture and lentivirus
  • Proteomics and protein-protein interaction
  • Gene regulation and transcriptomic analysis
  • ChIP
  • CRISPR/Cas9 technology
  • Luciferase reporter assays
  • Molecular biology techniques such as DNA isolation, qPCR, cloning, etc.
  • Cell cycle and cell death pathways analysis
  • Microscopy and immunofluorescence

Experience in the following areas will be considered as strong assets:

  • Post-translational modifications by ubiquitin and ubiquitin-like molecules
  • DNA damage response
  • Oxidative stress and antioxidant response
  • Flow cytometry

Experience in the following areas will be considered as additional merits:

  • In-vivo experience with mice
