The Department of Laboratory Medicine consists of seven divisions and is active in many areas of Laboratory Medicine. The department is involved in advanced basic scientific, clinical and epidemiological research. The department is one of the largest departments of education at Karolinska Institutet.

Division

The Clinical Research Centre (CRC), where the project will be carried out is a common resource for researchers from different departments at Karolinska Institutet and/or clinics at Karolinska University Hospital. The major part of the clinical research at Karolinska University Hospital is performed at CRC, presently comprising some 20 research groups with over 150 employees. The Centre provides the facilities needed for cell- and molecular biology research and the possibility to perform research in vivo.

EL Andaloussi’s research group works primarily on development of engineered extracellular vesicles, in particular exosomes, as therapeutic delivery vehicles for various biologics. The lab has different on-going projects related to the exosome field with a wide number of close collaborations with labs both within and outside Karolinska Institutet. Most of the projects are focusing on development of engineered exosomes for targeted delivery of protein therapeutics to treat various inflammatory conditions.

Duties

The project will focus on developing bioengineered extracellular vesicles aimed for treatment of inflammation. Most of the research will be conducted utilizing cell lines and animal models. The first part of the work will involve molecular cloning, transduction of cells and subsequent purification of engineered exosomes harbouring therapeutic proteins or RNA. Here, it will be important to phenotype EVs both on bulk mass basis as well as with single vesicle imaging. The second part involves assessing the uptake and biodistribution of these EVs in vitro and in vivo and to determine their biological activity in cell models as well as in disease animal models.

Prospective candidates should be highly motivated with the ability to work independently and as part of a team. Importantly, the applicant should be able to coordinate the work of other students and post docs in the lab.

Entry requirements and qualifications

The applicant should be a scientist with completed postdoctoral stints with a solid background in molecular biology or biotechnology and immunology. Practical lab experience is essential, especially in the following areas: cloning, expression and purification of recombinant proteins, fluorescent/confocal microscopy (ICC and IHC), protein analysis (Western blot, ELISA and IP), flow cytometry, cell culture work (knock-in and –out, establishing cell lines and cell assays), purification and quantification of extracellular vesicles, RNA expression analysis (rtPCR, qPCR and ddPCR).

The applicant should have previous experience from work with exosomes and exosome phenotyping using flow cytometry. Felasa certificate or equivalent animal experimentation experience for animal work is a prerequisite. In addition, excellent proficiency in both spoken and written English is highly desired.
