Karolinska Institutet, the Department of Medicine, Huddinge

Do you want to contribute to top quality medical research?

Department of Medicine (MedH)
The department of Medicine, Huddinge conducts graduate and post-graduate education, as well as research, within a wide range of the medical field, covering all internal medicine specialties, infectious diseases and dermatology. The department consists of seven units with a total of nearly 60 research groups. Our ambition is to be the leading department for medical research and education.

The group
We are a team of several research engineers, as well as senior scientists working together in a core unit where we define cells though their transcriptome. Our main technological focus is single cell sequencing using Smart-seq2 chemistry and the commercially available 10x genomics single cell solution. Our working group is made up of a mix of complementary skills in order to quickly and effectively drive research projects forward. Our projects have a different tissue focus, including vasculature, kidney, liver, heart and pancreas.

Duties
The post of the Research Engineer includes the following tasks:

  1. General day-to-day management of a core unit where a lot of projects are running in parallel,i.e. introducing new users to the specialized equipment, data sheet access management, reagent ordering and sample tracking.
  1. Preparation of 384-well plates containing RNA-lysis buffer. We are looking for a person who is very skilled at handling low volume (nanoliter range) dispensing using an Integra ViaFlo.
  2. Handling of single cell RNA extracts in 384-well plates and organizing the acquisition, tracking and progress of a large number of plates. Great organization and sample management skills are a merit.
  3. Conversion of RNA to cDNA using automated liquid handling robots (Agilent Bravo and LabCyte Echo 525) and PCR reactions. Enzymatic fragmentation and tagging of cDNA fragments for Illumina sequencing (HiSeq3000).
  4. Magnetic bead-based nucleotide isolations in 384 well plates, both manually and automated using the liquid handling robots described in the previous point.
  5. Quality control on cDNA conversion and sequencing library preparation using automatic microfluidic gel electrophoresis (Agilent Bioanalyzer).
  6. Clustering of cDNA libraries on an Illumina cBot2 for sequencing on an Illumina HiSeq3000.

Experience and competence
We are looking for an experienced person with extensive technical knowledge in the field of high- throughput liquid handling. The applicant should have previous experience with liquid handlers and should be proficient in working with sub-microliter volumes. Prior knowledge with sequencers or sequencing data is a merit. A large portion of the work will involve handling of many projects in a very dynamic laboratory, so skills in laboratory and project management are of large merit. As the research engineer will also be functioning as the main contact person for collaborative projects, excellent communication skills (both orally and written) are of the utmost importance. Prior experience with the abovementioned equipment is a strong merit for selection.

Qualifications
We are looking for a Research Engineer who is able to work dynamically at MedH, an international and multidisciplinary research department. Excellent knowledge of the English language is of utmost importance. Personal suitability will be a strong selection criterion. The applicant will be assessed on basis of technical competence, as well as past experience.
