This postdoctoral project relates to autoreactive T-cells identified in multiple sclerosis with a new immunological method. It is mainly comprised of two parts:
1. Deeper characterization of autoreactive T-cells specific for the novel autoantigens from patient and control samples using a combination of the established methods as well as T-cell proliferation assays and flow cytometry analysis. The exact methods regarding the proliferation and FACS will have to be, at least partly, developed during the project. The goal will be both on identifying potential MS-selective T-cell phenotypes such as CD4+, CD8+, Th1/2/17, effector-/memory-cells and cytokine/chemokines and receptors, as well as confirm previous FluoroSpot results regarding autoreactive T-cell frequencies.
2. Investigate the autoreactive profile of CNS-infiltrating T-cells. In these experiments, previous established FluoroSpot methods are not adequate as the antigen-presenting cell population is essentially absent in samples form the CSF. Modification of existing methods will have to be performed in order to effectively be able to analyze these types of cells.
The employment is for two years, with the possibility of an extension of two years.
Qualified to be employed as a postdoctor is one who has obtained a doctorate or has equivalent scientific competence. Applicants who have not completed a doctorate at the end of the application period may also apply, provided that all requirements for a completed degree are met before the (intended) date of employment. This must be substantiated by the applicant's main supervisor, director or equivalent.
