The Department of Neuroscience includes several research groups working on different aspects of nervous system function, both under normal conditions and in various disease states. Research projects include a broad range of studies on the molecular and cellular levels, networks of neurons, and the organization of the nervous system as a whole. The Department has a flat organization with 40 research group leaders, of which 24 are full professors, and 65 PhD students.

Teaching represents a fundamental activity at the Department, which participates in several undergraduate courses and postgraduate education programs. Each year approximately 15 students receive their PhD from our Department and many of our graduates have gone onto distinguished careers in the academic or private sector.

The Falk laboratory uses reprogramming, healthy and patient-specific iPS cells, and neural differentiation to derive cellular models in 2D and 3D of human brain development. In these models we study cellular and molecular mechanisms involved in healthy and diseased neural development. We are investigating phenotypes in the models using cellular assays as migration, proliferation, differentiation, imaging, flow cytometry, neuronal functionality in addition to bulk or single cell transcriptomics. We are also using genome editing strategies for correcting or introducing mutations in iPS cells for disease modelling. Our overall goal is to understand the molecular and cellular consequences of the emerging genomic findings for neurodevelopmental and neuropsychiatric disorders.

Description of the group
iPS core is a human cell facility, (www.ipscore.se), located at Karolinska Institutet in Solna. The facility was founded in 2013 by associate professor Anna Falk. The focus of the cell facility is culturing of human pluripotent stem cells, so called human induced pluripotent cells (iPS cells) and neuronal stem cells derived thereof. The services are aiming at deriving standardized cell cultures, cell culture advice and protocols for preclinical research.

Area of responsibilities/duties
Majority of the work will be advanced cell culturing (iPS cells and neural cells), and characterization of the generated cells. The work includes the whole reprogramming process from establishment of differentiated cells (for example fibroblast cells from skin biopsies) to reprogramming and expansion, characterisation and cryopreservation of established pluripotent stem cells cultures. Hands on cell culture demonstrations for researchers. Actively contribute to process development and standardization of existing and new processes. Contribute to a high standard of services and documentation.

Examples of specific duties:
Culture and cryopreservation of human iPS and rarely ES cells.
Derivation of human fibroblast cells from skin biopsies
Reprogramming of human fibroblast cells
Characterisation of iPS cells using immunocytochemistry, flow cytometry and quantitative RT-PCR.
Differention of stem cells to neuronal stem cells and other cell types.
Preparation of DNA and RNA
Ordering and inventory of medium and consumables
Documentation, electronic lab book
Medium preparation and factor aliquotation
Hand on cell culture training for researcher/ cell customers
Arrange with MTA-documents and cell shipments to customers
GMP cell production might be a part of the cell culture duties

Eligibility/Qualifications

The applicant should have an education in biology or equivalent.

Documented experience of culturing (with and without feeder cells) and cryopreservation of human ES and iPS cells is required.

Basic experience in molecular biology, immunocytochemistry, microscopy and flow cytometry is preferable. We have high demands on flexibility and accuracy since the stem cells are very sensitive.

Most of the work is advanced and labour intense cell culture with much routine and there is no opportunity to work on individual research projects.

Good ability to work in teams and to easily pick up new methods is a requirement. The applicant should be fluent in English, with good communication and writing skills since the working environment is international.

Personal qualities will be of great importance.
