The Chemical Proteomics Core Facility of Karolinska Institutet (ChemProt), Department of Medical Biochemistry and Biophysics, announces a staff position for Research Specialist in Chemical Proteomics and HDX-MS. ChemProt operates at Biomedicum and is also the specialized Chemical Proteomics Unit for two national infrastructures, Science for Life Laboratories (SciLifeLab) and the Swedish National Infrastructure for Biological Mass Spectrometry (BioMS). Dr. Massimiliano Gaetani is heading this Unit within the Division of Physiological Chemistry I, led by prof. Roman Zubarev.

The candidate will be key in the unique expertise present at ChemProt as well as advanced technical experience in a comprehensive and well-equipped laboratory. The last is multidisciplinary and includes: equipment for cell culture, ultracentrifuge, UPLC for off-line fractionation, and nine high resolution LC-MS Orbitrap systems, including the multiple and recently acquired systems of Fusion Lumos and Exploris480, Hydrogen Deuterium Exchange.

Description of the subject area

The Chemical Proteomics Facility / Unit is focused on providing full support to perform comprehensive support to research projects in need of:

  1. A) Liquid Chromatography coupled to Mass Spectrometry (LC-MS)-based proteomics to deconvolute how active molecules work in cells in terms of targets and action mechanism. Support to research projects is provided with coordinated service at including cell culturing treatment of cell cultures and lysates with molecule of interest, proteomics sample preparation, LC-MS analysis and bioinformatics data processing.
  2. B) Hydrogen/Deuterium exchange mass spectrometry (HDX-MS) for elucidation of protein binding interface is also a provided as service at the national level with specialization on using potentially therapeutic small-molecules and antibodies (epitope mapping) as ligands.

Both of these areas are expected to be covered by the winning candidate for this position of Research Specialist in Chemical Proteomics and HDX-MS.

Your mission

The work requires interaction with the staff and research personnel of the Chemical proteomics Unit and the rest of the Chemistry I Division. The duties include:

  • To perform chemical proteomics including all steps along the whole pipeline of work, including cell culture, drug treatments, proteomics processing procedures and final LC-MS analysis, according to the protocols at ChemProt
  • To perform the recently developed PISA assay (Gaetani M. et al. 2019), as well as multiplexed integrated approaches of expression proteomics, RedOx proteomics, and others, as assigned at ChemProt
  • To perform Hydrogen-deuterium exchange mass spectrometry (HDX-MS)
  • To perform data analysis of Chemical Proteomics and HDX-MS, including programming and using R scripts, and using specific software tools for chemical proteomics and HDX-MS
  • To participate and perform independently method development in both Chemical Proteomics and HDX-MS
  • To assist in instrumentation and lab maintenance relatively to the performed activities, including LC, MS and HDX PAL systems
  • To write SOPs, inventories, and analysis reports
  • To report directly to the Head of the Chemical Proteomics Unit on all the above.

Eligibility requirements

To be eligible for employment as a Research Specialist in Chemical Proteomics and HDX-MS, in addition to holding a Degree of Doctor, PhD, or having equivalent scientific expertise, the applicant must have demonstrated research expertise as well as have documented research experience after defending their doctoral dissertation.

The ideal candidate must have

  • prior experience with HPLC, UPLC, MS and HDX-MS PAL systems.
  • prior experience in quantitative proteomics, chemical proteomics, and integration with expression proteomics, with analysis at protein and peptide level
  • knowledge of nanoLC-MS using Orbitrap Q-Exactive series and Tribrid Lumos Series instruments
  • knowledge of capillary HOLC and peptide sample fractionation
  • experience with sample preparation in proteomics tasks
  • experience with bacterial and mammalian cell culture

A service-oriented personality, good inter-personal skills and the ability to efficiently work in an international research environment are absolutely required.

The candidate should be keen to learn and capture from potential professional training on HDX-MS, to performed on site and/or abroad (visits with duration of up to several weeks to the best labs).

As data analysis is key, bioinformatics and computer skills must include experience in programming on at least one computer language, and the ability (alternatively, desire to quickly learn) programming in R. 

English is the working language; good command of it in oral and written communications is a must.
