Division

The Department of Laboratory of Medicine, located in Campus Flemingsberg, is one of the Karolinska Institutet’s largest departments with 6 research divisions including the Division of Biomolecular and Cellular Medicine (BCM)(former Clinical Research Center), where the project will be carried out. BCM provides the facilities needed for cell- and molecular biology research and the possibility to perform research in vivo. EL Andaloussi’s research group works primarily on development of therapeutic delivery vehicles for various biologics. The lab has different ongoing projects with a wide number of close collaborations with labs both within and outside Karolinska Institutet. Most of the projects are focusing on development of engineered exosomes and peptide nanoparticles for targeted delivery of RNA- and protein therapeutics to treat various inflammatory conditions.

Your mission

The project will focus on the structural and biological characterization of the Immune System Released Activating Agent (ISRAA) protein. The ISRAA protein was first isolated in mice by using fluorescent differential display technology. The protein consists of 125 amino acids and the recombinant expressed protein revealed a 15 kDa molecule with biological activities similar to the cultured supernatants of splenocytes obtained directly from parasite-inoculated animals. In addition, antibodies raised against the protein blocked the activities and recognized a band in the active supernatant with the same molecular mass as the protein.

The literature suggests that ISRAA acts as a soluble ligand through cell surface receptor activation as well as an intracellular adaptor molecule. No receptor has so far been found. Neither is it clear how the protein can be active both extra- and intracellularly. The project aims at characterizing the structural and biological basis for ISRAAs actions using biological, chemical and biophysical methods.

Prospective candidates should be highly motivated with the ability to work independently and as part of a team. Importantly, the applicant should be able to coordinate the work of other students and post docs in the lab as well as cooperate with collaborative partners outside the lab.

Your profile

The applicant should be a scientist with completed postdoctoral stints and several years of work experience, with a solid background in biochemistry, molecular biology or biotechnology. Practical lab experience in protein characterization and cellular import is essential, especially in the following areas: Peptide synthesis, (HP)LC-purification and analysis, cell culture, receptor-ligand binding, CD-spectroscopy, fluorescence microscopy, cellular uptake assays and assay development. In addition, excellent proficiency in both spoken and written English is highly desired.
