Department of Medicine, Solna, Respiratory medicine division

The Department of Medicine, Solna, is one of the largest departments at Karolinska Institutet. The department consists of several divisions that are separated into different research groups. The core activities of the department are research, doctoral education and undergraduate education for medical and biomedical students. The research has three main profiles: allergy, cardiovascular medicine and inflammatory diseases.

Division
The Respiratory Medicine division at Karolinska Institutet, one of the divisions at the department, is now recruiting a Postdoctoral Researcher to investigate the mechanisms involved in complex respiratory diseases, with specific emphasis on pulmonary inflammation, and in particular in the inflammatory disease sarcoidosis. The successful applicant will work on cutting-edge projects in collaboration with international groups in the field.

Backgrund
We are looking for a post-doc to our Lung Research Lab, located in Bioclinicum, karolinska universityhospital, where we share the floor and equipment with the Division of Clinical Immunology. The lung research division consists of 4 research groups headed by J Grunewald, A Linden, M Sköld and Å Wheelock, respectively, in total consisting of about 15 individuals. The present position is for the Grunewald group.

Prodject
The applicant will focus on sarcoidosis, an inflammatory and granulomatous disease of unknown etiology, a field in which the group is internationally recognized. Our goal is to identify the antigen (or antigens) causing sarcoidosis, and/or to reveal markers of disease to improve e.g. prognostic accuracy.
In order to identify disease-associated antigens we have developed and refined the “HLA-peptide” elution technique, which has been used to identify peptides eluted from HLA-molecules, in our project derived from macrophages in sarcoidosis patients. The cells are obtained from the airways through bronchoscopy and bronchoalveolar lavage (BAL).
The goal is to further develop the HLA-peptide technique and subsequently apply it on already collected BAL cells of sarcoidosis patients, and eventually of patients suffering from other diseases.

Duties
The applicant will need experience from working with peptides and proteins, e.g. in cell stimulation test, and to have competence in protein chemistry and bioinformatics. An interest and competence in techniques relevant for the HLA-peptide elution technique is necessary, and knowledge on mass spectrometry (MS) is an advantage. Further, to be familiar with working with cells, using tissue culture, cell isolation, flow cytometry e.t.c. is a plus.
The applicant will eventually run the HLA-peptide technique with patient samples. If successful, the HLA-peptide-elution technique may generate a new way of identifying “peptide-patterns” that may associate with disease and/or prognosis.

Entry requirements
We need an independent post-doc with competence in protein chemistry, bioinformatics and immunology (in that order). Knowledge of appropriate laboratory methods is necessary. The applicant also need skills and interest to sum up experiments, interpret results and plan new experiments, as well as in writing articles. Qualified to be employed as a postdoctor is one who has obtained a doctorate or has equivalent scientific competence.
